Rachel, I have not been involved in doing this at least not as a medical test performed in a clinic. Duke has a pilot program for nurses and you may want to reach out to Carol Epling there for her insights.

I attended an excellent presentation at a work comp conference by an ergonomist named Jim Briggs who really crystallized the legal do's and don'ts of post offer physical ability testing. However by "skills" day what you are asking sounds more like something the hiring department might do regularly for existing employees, not post offer. That is a more common practice than post offer physical ability testing, and as long as it's not done by medical personnel and everyone goes through it, and it's a reasonable approximation of the job duties and demands, I don't see a reason they couldn't do it. I would resist turning that into a medical test though.
